The game is the fact that he deliberately made errors in his writing to see if we noticed them. He made about 100 of them.
Everybody has their own variation of defining an error. Some errors stand out to some people while others don't even notice them. Errors exist in so many places, that it is impossible for every person to identify the same ones and respond to them the same way.
He located errors in the reader locating errors.
He wanted to make the point that nobody seemed to notice that E.B. White made an error. He is amazed by the fact that the guy that notices errors in others writing didn't notice it in his own.
